Fragment片段库

基于片段的药物设计(FBDD)的理论基础是挑选有利的片段组合或延伸得到新药物分子,获得高活性的候选药物,其本质不是筛选已知化合物的类似物,而是培育和设计药物。FBDD方法的前提是建立高品质的片段库,片段分子往往需要满足“三规则”:分子量小于300,氢键供体和受体不超过3(未得到广泛认可),可旋转键数目不超过3,cLogP小于3,此外良好的化学空间多样性也是重要的考察内容。
