It was generally thought that hepatic macrophage is the target of overloaded iron to induce inflammation. However, in our submitted manuscript, we showed that, both in vitro and in vivo, iron-overload leads to inflammatory responses directly through the hepatocytes.

Using a genetically modified mouse model, previous studies indicated that microRNA-122 expression is significantly  decreased upon iron-overload. However, the molecular mechanism is unclear. In our submitted manuscript, we concluded that during the process of transfusional iron-overload in genetically unchanged  mice, only HNF4, but not other upstream regulators, is significantly  reduced, which in sum regulates microRNA-122 expression.

Taken together, our findings elucidate previously-unknown mechanism of both cellular and molecular targets during iron metabolism in the liver. We believe that our findings are informative for developing preventative clinical therapies.
