Strategy for discovery and validation of DC and monocyte subtypes
To determine the subtypes of DCs and monocytes in human blood, we developed an experimental and computational strategy to
(i) perform single-cell RNA sequencing on DCs and monocytes derived from a single healthy individual; 单细胞测序,获得健康组织数据
(ii) identify clusters of cells that are similar to each other; 聚类分析
(iii) find discriminative markers per cluster; 找出区分每个类别的marker基因
(iv) prospectively isolate cells corresponding to key clusters using newly identified surface markers; 利用新鉴定的marker基因将一些潜在的细胞亚型从大类中分出来
(v) validate the identity of the sorted cells using scRNA-seq;
利用scRNA-seq鉴定挑选的细胞特性
(vi) confirm the existence of these cell types in up to 10 independent healthy individuals; and 确认这些细胞类型普遍存在于多个样品
(vii) perform functional analyses for selected cell types. 指定细胞类型的功能分析
个人认为最艰难的还是第3和第4步,不把文献库翻箱倒柜..把该组织和各个细胞以及它们的marker基因搞懂的话..要出好结果还是很难的....
